Kits
There are three kits available, all
relying on using standard EDC chemistry and carboxyl bonds
to bind to primary amines. The first kit comes
complete with carboxyl-polymer conjugated to a customer chosen spherical gold
nanoparticle, EDC chemicals, and instructions to conjugate
an included IgG to our gold nanoparticles. This kit
includes discussions on theory, background, purification,
and other important principles involved. The second
kit is for the more advanced user. This one includes
the EDC chemicals and gold nanoparticles for use in conjugations
involving user supplied antibodies or dna. It does not
include instructions or IgG - it is recommended that the
customer purchase Kit 1 first. Kit 3 is for the most
experienced user that has already purchased our gold
nanoparticles in 250 OD-mL volumes and need the EDC
chemicals for their conjugation.
Part number 21-PC-KIT1
This is a kit to instruct the
conjugation of carboxyl-polymer spherical gold nanoparticles
(SGNPs) to any primary amine using EDC chemistry. This kit
includes IgG as the conjugation
target. The customer specifies the gold nanoparticle
size desired (5-100nm).
EDC is a carboxyl and amine-reactive
zero-length crosslinker. EDC reacts with a carboxyl group
first and forms an amine reactive O-acylisourea
intermediate that quickly reacts with an amino group to form
an amide bond and release of an isourea by-product (see the
Additional Information Section). The intermediate is
unstable in aqueous solutions, and therefore, two step
conjugation procedures require N-hydroxysuccinimide
for stabilization. Failure to react with an
amine will result in hydrolysis of the intermediate,
regeneration of the carboxyl and release of an N-substituted
urea. A side reaction is the formation of an N-acylurea,
which is usually restricted to carboxyls located in
hydrophobic regions of proteins.
Part number 21-PC-KIT2
This kit provides all the chemicals
necessary to perform EDC chemistry on our carboxyl-polymer
spheres. This is the same as KIT1 but without the IgG.
It is targeted to those that are experienced in KIT1 but now
want to attach our gold nanoparticles to their own
antibodies or other target with a primary amine. Both
our KIT1 and KIT2 are for 50 OD-mLs of gold nanoparticles.
Part number 21-PC-KIT3
This kit provides all the chemicals
necessary to conjugate 250
OD-mLs of gold nanoparticles. Unlike KIT2, it does not
contain gold nanoparticles.
Custom Formulation
These kits are based on EDC chemistry
activating carboxyl terminated polymer conjugations so as to
attach to any primary amine.
